Cognitive disorders and symptoms are key features of many mental neurological diseases, with a large spectrum impaired domains. Because their possible evolution detrimental functioning impact, they major pharmacological target for both symptomatic disease-modifier drugs, while few cognitive enhancers have been marketed an insufficient efficiency. It explains the need to model these beyond modelization or diseases themselves. According experimental strategy used induce impairment, three categories models identified: neurotransmission-driven models; pathophysiology-driven environment-driven models. These reflect different levels integration endogenous exogenous mechanisms underlying in humans. Their comprehensive knowledge illustration modulation could help propose renewing drug development central nervous system (CNS) field at time when academic industrial invest seems be declining despite medical social burden brain diseases.
